Bottom line
The sleeper story is Vertex's unprecedented visibility into gene therapy revenue through their patient journey mapping-Duncan's 'very strong visibility' claim is rare transparency that makes the $500M non-CF target more credible than typical biotech promises. The obvious headline is Povi's stellar Phase III data, but the real constraint is CF growth maturation-Reshma's admission that there's 'very little unmet need...it's getting really, really tough' for further CF innovation. The most revealing quote: management doubled down on renal being their 'fourth commercial pillar' while simultaneously acknowledging CF innovation limits. My view changes if Q2 CF growth doesn't stabilize above 5% or if non-CF revenue run-rate doesn't hit $125M quarterly by Q3.
